Research

Glycosylation is a well-known posttranslational modification that functions in a variety of biological roles ranging from the stabilization of protein structure to the regulation of cell surface properties. The general interest of our Lab is to understand the molecular mechanism of this biological process in eukaryotic cells and further explore the functions of various glycans. We mainly study on Asparagine (N)-linked glycan and Glycosylphosphatidylinositol (GPI)-anchored modification pathways, using a palette of technologies including MS/LC-MS analysis, protein imaging, genetic screens, CRISPR/Cas9 & targeted genome editing techniques, in vitro chemo-enzyme synthesis, and so on. In addition, we are developing a cell expression system for production of pharmaceutical proteins, and also challenging the bioprocess for functional sugar production and its industrial application. Several research topics are listed below:

1. Molecular mechanism of N-glycosylation in eukaryotic cells and its in vitro assembly.

2. Analysis of particle uptake and endocytic pathways in mammalian cells and its applications.

3. “Traffickome” of glycoproteins and biopharmaceuticals (Integrated analysis of protein glycosylation, folding and transport).

4. The function of sugars and biosynthesis of the sugar using microorganisms and related enzymes.

5. Development of efficient production of therapeutic proteins with homogeneous glycosylation system

6. Glycosylation in tumor and diseases, Bioinformatics and multi-Omics analysis of tumor biology
